Honorable mention: Venom

Tom Hardy’s Venom has spawned Sony’s own universe of Spider-Man characters, with the sequel being recently released to both critical and financial success. Both Hardy and Tom Holland have expressed interest on having their characters cross paths and it may occur in Spider-Man: No Way Home.

However, Eddie Brock only receives an honourable mention here as the Venom portrayed by Hardy is more of an anti-hero character than a straight villain. He has taken on both Riot and Carnage in separate films and has been known to team up with Spider-Man in recent comics.

The reveal of Spider-Man’s identity and Parker being framed for murder could see the two characters do battle, however it is hard to see Brock doing a full heel turn and joining the Sinister Six unless he is manipulated by Doctor Octopus or some other character who makes Venom believe they are the good guys.

Doctor Octopus

As confirmed by the actor, subsequent stills and the first full trailer, Doctor Otto Octavius (portrayed by Alfred Molina) from Sam Raimi’s Spider-Man 2 will be in Spider-Man: No Way Home. He has been the villain most featured in promotional materials for the film and seems to be a major thorn in Peter Parker’s side.

Doc Ock has been the leader of the Sinister Six in the majority of their appearances in both comic books and other media, with the same likely being the case in the team’s first live action appearance. Ock has the genius and the skillset to lead this team, with his experiences in Spider-Man 2 against Tobey Maguire’s Spidey showcasing that he knows what it takes to take on Spider-Man and defeat him (as he did in the train sequence).

Doc Ock will be a big threat and a cold and calculating leader for the Sinister Six, which will consist of some of Spider-Man’s biggest enemies from across the multiverse.
